Data from the cDNA microarrays described in "Mediator protein mutations that selectively abolish activated transcription" (Proc Natl. Acad. Sci. USA, 96, pp.000-000,1999) may be downloaded as text tab delimited files.
In these experiments, polyA mRNA from the wild-type strain MG106 was reverse transcribed in the presence of Cy5-dUTP (red), and polyA mRNA from the med2 deletion strain MG107 was reverse transcribed in the presence of Cy3-dUTP (green). Elements of the array that appear red correspond to genes transcribed less efficiently in med2 deletion mutant relative to the wild-type strain.
The data are sorted by red-to-green ratio (listed in the column N RATIO WT/DMED2). Genes transcribed least efficiently in the med2 deletion mutant relative to wild-type appear at the top of the list.
Note: Data from elements of the array with visible imperfections are not included.
The column headings of the data files are as follows:
NAME Six letter alphanumeric code from SGD for each open reading frame (ORF) on the arrays.
GENE Standardized format for the name of a genetic marker consisting of three letters (the gene symbol) followed by an integer (e.g. GAL4).
WT INT Intensity of the red fluorescent probe hybridized to a given ORF.
WT BKG Background correction factor for the red fluorescence.
N DMED2 SIG Normalized intensity of the green fluorescent probe hybridized to a given ORF. Normalization was performed using genomic DNA spotted onto the array which are expected to yield a ratio of 1 for net red signal/net green signal.
N DMED2 BKG Normalized background correction factor for the green fluorescence.
N RATIO WT/DMED2 Ratio of the net signal minus background for the wild-type vs med2 deletion mutant. (red to green ratio).
QUAD The arrays consist of 4 quadrants each having the dimensions 39 rows by 42 columns. The coordinates for the quadrant, row and column can be used to identify the spot on the array for a given ORF. The quadrants are numbered from one, two, three and four from the top left, top right, bottom left and bottom right, respectively.
ROW The rows of spots in each quadrant are numbered from top to bottom, 1 though 39.
COLUMN The columns of spots in each quadrant are numbered from left to right, 1 through 42.
DESC Description of the function of the protein encoded by the open reading frame, if known.
